Suppressor of Cytokine Signaling (SOCS) 1 is a key negative regulator of interferon (IFN), interleukin (IL)-12, and IL-2 family cytokine signaling.
“While the mechanisms that govern selective regulation by SOCS1 remain an enigma, one obvious possibility is that inhibition of signaling depends on exceeding a certain threshold in SOCS1 expression,” stated Professor Sandra Nicholson and Dr. Karen Doggett, from The Walter and Eliza Hall Institute, and coauthors. “Here we have characterized a new SOCS1 reporter mouse strain that expresses a Halo-tagged SOCS1 fusion protein under control of the endogenous Socs1 promoter elements. Cells from these mice were utilized to track SOCS1 expression and investigate the relationship between SOCS1 levels and inhibition of signaling.”
The investigators concluded that SOCS1 needs to exceed a threshold level to inhibit both IFN-gamma and GM-CSF signaling. Furthermore, “the ability of SOCS1 to cross-regulate GM-CSF signaling is directly related to the level of SOCS1 expression.”
