High Speed Volumetric Light Sheet Imaging of Awake Behaving Mouse Brain (VIDEO)

This movie shows a volume of data acquired using SCAPE at 10 volumes per second over a large field of view (800 micron width). Apical dendrites of layer five neurons expressing GCaMP can be seen in addition to vasculature labeled with intravenous Texas red dextran. Serial sections and dynamic sequences demonstrate that SCAPE can capture real-time 3-D dynamics in the superficial layers of the cortex with high resolution at over an order of magnitude faster speeds than two-photon microscopy using a low-cost 488 nm 30 mW laser. Additional published data identifies unique, spontaneous calcium dynamics in individual branches of 3-D dendrites in awake behaving mouse brain.
