Cpf1 off-target effects were avoided by delivering Cpf1 and CRISPR-RNA complex, rather than plasmids encoding Cpf1 and crRNA. The accuracy of genome engineering was enhanced In this study, the researchers demonstrated methods to efficiently deliver CRISPR-Cpf1 genome engineering molecules. The researchers delivered pre-assembled protein-RNA complex (right) rather than using plasmid DNA that encodes Cpf1 and RNA (left), and showed DNA cleavage exclusively at target site.
