Genome-wide CRISPR knockout screens to identify cellular regulators of LNP-based mRNA vaccines. mRNAs encoding Green Fluorescent Protein (GFP) were in vitro transcribed (IVT) and delivered to knockout cells using LNP. Cells with high or low GFP fluorescence were collected and sequenced to uncover cellular regulators of LNP-encapsulated mRNAs.
