The region highly conserved in land plants (yellow area) contains two Ca2+ binding sites (green area). Without Ca2+ binding, RBOH is not activated, and Ca2+ binding to the Ca2+ binding site is essential for reactive oxygen species (ROS) generation; phosphorylation of the two serine residues (in red) increases the binding affinity of Ca2+ (making Ca2+ easier to bind).
