Snake puzzle models of protein folding and amyloid fibril formation. Protein folding under Anfinsen's dogma leads to a unique functional native structure. Protein misfolding out of Anfinsen's dogma leads to amyloid structures. The two reactions are separated by the high free energy barrier of supersaturation. Once this barrier is broken by agitation or seeding, the equilibrium of folding and misfolding will be achieved.
