An Alternative Target for Cancer Therapy at the Ends of Chromosomes (VIDEO)

This movie shows that an ATR inhibitor induces numerous DNA breaks in ALT-positive cancer cells. ALT-positive U2OS osteosarcoma cells were treated with the ATR inhibitor VE-821 and analyzed by live time imaging. The green dots in the nuclei are foci of 53BP1, a marker of DNA breaks. The appearance of green dots in these cells demonstrates that ATR inhibition induced many DNA breaks in the genomes of these cells. This material relates to papers that appeared in the January 16, 2015 issue of Science, published by AAAS. The paper, Rachel Litman Flynn at Massachusetts General Hospital Cancer Center in Charlestown, MA, and colleagues was titled, 'Alternative lengthening of telomeres renders cancer cells hypersensitive to ATR inhibitors.'
