hMTH1 recognizes two oxidized dNTPs as shown in the 3-D structure (upper area of figure: hMTH1, ribbon; damaged dNTPs, sticks). The determining factor in the broad substrate specificity is differing protonation states between Asp-119 and Asp-120 (lower area of figure).
