TGFβ/ACTIVIN-SMAD2/3-EZH2 cooperates with OCT4 and NANOG in self-renewing hPSCs to keep CDKIs in an epigenetically poised state for rapid activation. Upon the initiation of endoderm differentiation SMAD2/3 switches its binding to SMYD2 and EOMES to induce CDKI expression that guides tissue formation through CDK inhibition.
